Biological Analysis

The technique of removing cells from the living environment and growing them under special and controlled conditions is called cell culture. In vitro tests are performed in laboratory conditions using a variety of cell lines. In vitro tests are rapid and animal-friendly. The biological effectiveness of any newly developed product and whether it has a toxic effect (cytotoxicity) on cells can be proven by cell culture studies.

In vitro cytotoxicity tests are measurement methods used in cell culture studies for the evaluation of agents that have drug characteristics or whose toxic profile is being investigated. With these tests, it is possible to analyse many substances in a short time and basic data can be obtained for in vivo animal and clinical trials. Although there are many cytotoxicity tests with different mechanisms (MTT, MTS, XTT, WST), two of the most widely used methods are MTT and WST. MTT is a colorimetric method used to measure cellular metabolic activity as an indicator of cell viability, proliferation, and cytotoxicity. It is widely used because it is fast, versatile, quantitative, and reproducible.

As MecNano Technologies, we do the following in vitro tests for your products with our Science Team:

  • Cytotoxicity Tests (MTT and WST)
    • 3T3 (mouse fibroblast cell line)
    • L929 (mouse fibroblast cell line)
  • Irritation test (for biocidal products in direct contact with the human body)
    • Skin irritation tests in biocidal products are performed with the “OECD Test Guideline 439 In Vitro Skin Irritation: Reconstituted Human Epidermis Test Method”.
    • Skin irritation tests and cytotoxicity tests in cosmetic products are performed with OECD Test Guidelines and TS EN ISO 10993.
  • Anti-Alzheimer's Efficacy Test
    • Amyloid Beta-induced SH-SY5Y (neuroblastoma cell line)

The analysis methods we use within the scope of cell culture studies are as follows:

ELISA

ELISA (Enzyme-linked immunosorbent assay) technique is a technique based on antigen and antibody complex, using enzymes for marking purposes. The ELISA technique results in a colored final product that is absorbed at a particular wavelength and can be correlated with the amount of substance present in the sample. A highly specific antibody and antigen interaction is the most critical component of this process. Depending on the combination of antigen and antibody, there are direct ELISA, indirect ELISA, sandwich ELISA and competitive ELISA.

As MecNano Technologies, we analyse the effects and mechanism of action of your products with sandwich ELISA at the end of in vitro cell culture studies with our Science Team.

PCR

Real-time polymerase chain reaction (Real-time PCR) is a PCR method that can give quantitative results by measuring the fluorescent signal that increases simultaneously with DNA amplification.

Real-time PCR technology is increasingly gaining importance in various fields favored by users, allowing for a more detailed analysis of the effects of active pharmaceutical agents at the cell level. The reason why it is preferred is that it is more sensitive, efficient, faster, and more productive in gene expression.

As MecNano Technologies, we analyse the detailed effects and mechanism of action of your products with real-time PCR at the end of in vitro cell culture studies with our Science Team.

Western blot

Western blot is a molecular biology technique that allows to analyse a specific protein in a cell or tissue. With this technique, the presence, size, concentration, and concentration changes of a protein in cells and tissues can be detected and the concentrations between different groups can be compared. Having high specificity and enabling the imaging of more than one protein at the same time provides a great advantage in protein expression studies.

As MecNano Technologies, we analyse the detailed effects and mechanism of action of your products with the Western blot technique at the end of in vitro cell culture studies with our Science Team.
